摘 要
海藻糖是一种非还原性二糖,理化性质稳定,无毒无害.在生物体内,它通常是响应环境变化的应激性产物,具有显著的保护性能,而且对离体生物大分子也具有良好的非特异性保护作用.这些特性引发了世界性的对海藻糖的研发热潮.本研究采用易于培养的莱茵衣藻为实验材料,使用Western印迹法检测在盐胁迫、热胁迫和干旱胁迫条件下衣藻中海藻糖水解酶表达情况,揭示不同胁迫下衣藻中海藻糖参与抗逆代谢的可能模式.实验结果表明:
42℃热休克处理两小时的衣藻中海藻糖水解酶蛋白表达量在处理开始的1小时即开始增加并达到最大,从第2小时开始逐渐下降,到第4至8小时达到稳定水平.
在75mM NaCl胁迫处理下,衣藻中海藻糖水解酶表达量在盐胁迫的第1小时明显增加并到达最大值,之后从胁迫的第2小时逐渐减弱,第4至8小时达到稳定水平.
在500mM山梨糖醇处理的干旱胁迫条件下衣藻海藻糖水解酶表达量胁迫处理后迅速增加,并在1小时左右达到最大值,之后有所下降,并在2小时至8小时处于稳定水平.
说明在上述三种胁迫条件下可能海藻糖代谢参与衣藻抗逆性反应的模式相似.

Abstract
Trehalose is a non-reduction of the sugar,physical and chemical properties and stability,non-toxic harmless.In the body,it usually is in response to environmental change the product of stress,a significant protective properties,but also for in vitro biological macromolecules also has a good non-specific protection.These features triggered a worldwide research and development boom of the trehalose.This study used easily cultured Chlamydomonas reinhardtii for the experimental materials,the use of Western blot test in salt stress,heat stress and drought stress conditions in Chlamydomonas trehalose enzyme expression,and reveal different stress Chlamydomonas participation in the anti-trehalose Inverse metabolism of possible models.The results showed that:
42 ℃ heat shock two hours of Chlamydomonas in trehalose enzyme protein expression in dealing with the beginning of the one hour that is beginning to increase and reach the most,from the first two hours and then a steady decline,to the first 4-8 hours to achieve the level of stability.
In 75 mM NaCl stress,Chlamydomonas trehalose enzyme in the expression of salt stress in the first one hour and reach maximum increased significantly,from stress after the first two hours gradually weakened,4-8 hours to achieve the level of stability.
In 500 mM sorbitol deal with the drought stress conditions Chlamydomonas trehalose hydrolase expression of stress after the rapid increase of around one hour and reached the maximum,after a decrease,and in 2-8 hours at a stable level.
In the three stress that under the conditions of trehalose metabolism may be involved in Chlamydomonas resistance pattern similar to the reaction.
