colleagues investigated the consequences of depleting Rad52 from cultured human
cells lacking BRCA2.They found a striking synthetic lethal effect on growth
and synergistic effects on DSB-induced homologous recombination and
radiation-induced chromosome abnormalities.Their observations support a model
in which Rad52 does indeed play a role in repair of DSBs and in overcoming
DNA-replication fork stalling,but that role is masked when BRCA2 is present.
The investigators propose that Rad52 cooperates with Rad51 in an alternative repair pathway that is
independent of BRCA2.
